Abstract

Obesity is a chronic metabolic disease characterized by excessive fat accumulation resulting from a long-term imbalance between energy intake and expenditure. This condition is closely associated with insulin resistance, chronic inflammation, dyslipidemia, elevated blood glucose, and increased total cholesterol. Magnesium, particularly magnesium glycinate with its high bioavailability and good gastrointestinal tolerability, has attracted considerable attention as a supportive nutritional therapy. This literature review aimed to analyze the effectiveness of magnesium glycinate supplementation on blood glucose and total cholesterol levels in obesity induced by a high-fat and high-fructose diet. This review was conducted by searching scientific articles from PubMed, Scopus, ScienceDirect, SpringerLink, and Google Scholar. Eligible studies were published between 2018 and 2025 and discussed obesity, high-fat high-fructose diet, magnesium glycinate, glucose metabolism, lipid metabolism, and experimental animal studies. Evidence indicates that magnesium glycinate improves insulin sensitivity by activating insulin receptors and GLUT-4 transporters, suppresses chronic inflammation through inhibition of the NF-κB pathway, and reduces oxidative stress. Furthermore, magnesium glycinate improves lipid metabolism by reducing cholesterol synthesis and increasing fatty acid oxidation, thereby contributing to reductions in blood glucose and total cholesterol levels in obesity models. Magnesium glycinate has considerable potential as a supportive therapy for obesity through improvement of glucose and lipid metabolism. Its high bioavailability makes it more effective than several other magnesium formulations in improving hyperglycemia and dyslipidemia.
